Abstract:
با توجه به اهمیت تشخیص افتراقی میکروارگانیسم های خانواده انتروباکتریاسه و ویبریوناسه به عنوان شایعترین عوامل ایجاد کننده اسهال و سایر بیماریهای عفونی و با توجه به مشکلات و معایب محیط های رایج مورد استفاده، این تحقیق به منظور طراحی و فرمولاسیون محیط کشت دو منظوره برای پرورش میکروارگانیسم های هر دو خانواده در یک محیط واحد طی سال های 77-1376 انجام گرفت. پژوهش حاضر به روش توصیفی (Descriptive) از نوع اکتشافی صورت پذیرفت. مواد شیمیایی پایه مورد نیاز برای رشد این گروه میکروارگانیسم ها بررسی شد و پس از تغییر در نسبت مواد شیمیایی اولیه (نمک، پپتون، قندهای مختلف و ...) و افزودن برخی مواد (فنیل آلانین، فریک آمونیوم سولفات و ...) هر کدام با هدفی خاص منجر به پیدایش فرمولاسیون محیط کشت جدیدی گردید. میکروارگانیسم های هر دو خانواده که اغلب از سوش های استاندارد از مراکز کلکسیون باکتریهای صنعتی، پزشکی و انستیتوپاستور تهیه شده و تعدادی از آنها نیز از سوش های غیر استاندارد بودند که هویت آنها از طریق کشت بر محیط های افتراقی متعدد به صورت دقیق اثبات گردیده بود، بر روی این محیط جدید مطابق با استانداردهای کنترل کیفی ATCC پرورش داده شد و این کار سه بار تکرار گردید. سپس، ویژگیهای کلنی های رشد یافته به طور کامل بررسی و براساس آن یک الگوریتم تشخیصی پیشنهادی ارایه شد. هزینه این محیط برابر با محیط های مصرفی رایج بوده و می تواند در بخشهای میکروب شناسی تمامی آزمایشگاه ها مورد استفاده قرار گیرد. استفاده تجربی از این محیط جدید در برابر روشهای رایج فعلی توصیه می گردد.
Considering the importance of differentially diagnosing microorganisms of the Enterobacteriacea and Vibrionacea families as the most common etiologic agents of diarrhea and other infectious diseases, and due to the difficulties and pitfalls of using common culture media, this study was performed to design and formulate a dual-purpose culture medium to grow microorganisms of both families in a single medium. In this descriptive study, the basic chemical substances needed for the growth of these microorganisms were studied, and after changing ratios of the primary chemical substances (e.g. salt, peptones, carbohydrates, etc.) and adding other substances (e.g. phenylalanine, sulfate) with a specific aim each, a new culture medium was formulated. Microorganisms of both families - mostly standard strains of the Pasteur Institute and industrial and medical bacteria collection centers, and some non-standard strains precisely identified by culturing on several differential media - were cultured three times on the new medium according to ATCC quality control standards. Characteristics of the grown colonies were thoroughly studied and a diagnostic algorithm was proposed. The cost of the new medium is equal to the cost of common media, and it may be used in microbiology departments of any laboratory. Experimental use of the new medium is recommended on a par to other currently used media.
