چکیده:
سابقه و هدف: بافت چربی می تواند فاکتور نکروز دهنده توموری آلفا تولید نماید. سطوح افزایش یافته این سایتوکین با چاقی و مقاومت به انسولین در ارتباط است. هدف این مطالعه، بررسی ارتباط بین پلی مورفیسم های G-308A وG-238A با چاقی در جمعیت ایرانی است.
مواد و روشها: افراد شرکت کننده در مطالعه قند و لپید تهران در دو گروه سنی زیر 18 و بالای 18 سال دسته بندی شدند. افراد بالای 18سال، بر اساس نمایه توده بدنی در سه گروه و افراد زیر 18سال، براساس صدک BMI در دو گروه قرار گرفتند. در مجموع244 نفر آنها جهت بررسی جایگاه 308- و 239 نفرشان جهت بررسی جایگاه 238- انتخاب شدند. بررسی پلی مرفیسم های مذکور با استفاده از تکنیک های PCR و RFLP انجام شد.
یافته ها: فراوانی الل های TNF-αاز قانون هاردی واینبرگ تبعیت می کرد و هیچ یک از دو پلی مورفیسم مذکور، ارتباط معنی داری با نمایه توده بدنی نداشت.
نتیجه گیری: داده های این مطالعه حاکی از عدم وجود ارتباط بین پلی مورفیسم های G-308Aو G-238A ژن TNF-α با چاقی بوده و بنابراین احتمالا این پلی مورفیسم ها نقشی در افزایش خطر بروز چاقی و بیماریهای مرتبط در جمعیت ایرانی ندارند.
Background and Aim: Tumor necrosis factor-α (TNF-α) is expressed primarily in adipocytes and elevated levels of this cytokine have been linked to obesity and insulin resistance. Therefore، we examined the relationship between the G-308A and G-238A polymorphisms of TNF-α gene promoter variants and obesity in an Iranian population.
Materials and Methods: Subjects of the Tehran Lipid and Glucose Study classified into two age groups under and above 18. Adults classified in three groups according to their body mass index and less than 18 years old subjects classified in two groups (under 85th percentile for their age and sex and above 85th percentile). 244 persons were selected to examine -308 site and 239 persons were selected to examine -238 site. The mentioned polymorphisms were examined with PCR and RFLP methods.
Results: The allele frequency of TNF-α polymorphism was in the Hardy Weinberg equilibrium and there was no relation between BMI and the frequency of this allele.
Conclusion No association between G-308A and G-238A TNF-α promoter polymorphisms and obesity could probably indicate that it is not an important risk factor for obesity and consequently for cardiovascular disease
