چکیده:
صرع از جمله اختلالات مزمن سیستم اعصاب مرکزی است، علیرغم تحقیقات گسترده ای که بر روی این بیماری صورت می گیرد و وجود داروها و روشهای متعدد در درمان آن، پاتوفیزیولوژی این بیماری هنوز هم بخوبی شناخته نشده است. علاوه بر این درصد قابل توجهی از بیماران مصروع نسبت به داروهای موجود ضد صرع مقاوم می باشند. از طرف دیگر در کتب طبی سنتی معتبر، گزارشهای متعددی در مورد اثرات ضد تشنجی برخی گیاهان از جمله زیره ایرانی (Bunium Persicum) وجود دارد. بررسیهای اولیه آزمایشگاهی، اثرات ضد تشنجی اسانس میوه زیره را در مدل های تشنجی ماکزیمال الکتروشوک و پنتیلن تترازول حاد تایید کرده است از این رو پیشبرد تحقیق روی این گیاه با استفاده از مدل های صرعی آزمایشگاهی پیشرفته تر ضروری به نظر می رسد. در تحقیق حاضر با استفاده از مدل «کیندلینگ-پنتیلن تترازول حاد» اثرات احتمالی ضد تشنجی اسانس زیره مورد بررسی قرار گرفت. نتایج حاصل از تزریق داخل صفاقی مقادیر 0.25-1 mI/kg از اسانس و اتوسوکسیمید 300mg/kg (کنترل مثبت) به فواصل زمانی 30 و 60 دقیقه قبل از تزریق پنتیلن تترازول حاد درموش های کیندل شده، نشان می دهد که اسانس زیره با مقادیر استفاده شده، در این مدل تشنجی بی اثر است. علت این امر احتمالا ناشی از تفاوت های موجود در علل بروز و نوع تشنج در مدل کیندلینگ-پنتیلن تترازول حاد، نسبت به مدل های ماکزیمال الکتروشوک و پنتیلن تترازول حاد می باشد.
Despite extensive research on epilepsy and development of various drugs and treatment methods for this chronic disorder of the central nervous system, the pathophysiology of the disease remains unclear. In addition, a significant percentage of epileptics are resistant to available antiepileptic drugs. On the other hand, many reports on the anticonvulsant effects of plants such as Persian cumin (Bunium persicum) have been recorded in the authoritative texts of traditional medicine. Preliminary laboratory studies using the maximal electroshock (MES) and acute pentylenetetrazol (PTZ) seizure models have substantiated the antiepileptic effects of cumin extract. Therefore, more extensive studies on this plant through the use of more advanced epilepsy lab models seem necessary. In this study, we investigated the possible antiepileptic effects of cumin extract using the PTZ-kindling model. Results of intraperitoneal injection of 0.25-1 ml/kg extract and 300 mg/kg ethosuximide (as positive controls), 30 and 60 minutes before injection of PTZ in kindled rats, showed that this dose of cumin extract has no effect in this seizure model.
