چکیده:
زمینه وهدف: تحقیق حاضر با هدف بررسی اثر تعاملی تمرینهوازی و مصرف مکمل کورکومین بر بیانژنNRF-1 و NRF-2 میتوکندریایی کاردیومیوسیت موشهای صحرایی نر مدل سکته قلبی انجام شد. روش پژوهش: 32 سر موش صحرایی نر به صورت تصادفی به چهار گروه کنترل، تمرینهوازی، مصرف مکمل و تمرینهوازی + مصرف مکمل تقسیم شدند. گروههای تمرین و تمرین + مکمل به مدت 8 هفته در برنامه تمرینهوازی شرکت کردند. تمرین روی نوارگردان الکترونیکی هوشمند حیوانی به صورت 5 روز در هفته انجام شد و در این زمان گروه کنترل و مصرف مکمل برنامه تمرینی نداشتند. همچنین، مکمل کورکومین پنج روز در هفته و به صورت گاواژ به گروههای مکمل و تمرین + مکمل داده شد. پس از جراحی، بیانژنهایNRF-1 و NRF-2 کاردیومیوسیت با استفاده از روش Real-time PCRبهدست آمد. برای تجزیه و تحلیل ﺩﺍﺩهها ﺁﺯﻣﻮنهای شاپیرو-ویلک و تحلیل واریانس دوطرفه مورد استفاده قرار گرفت. یافتهها: هر دو متغیر تمرین و مکمل کورکومین به صورت معنیداری بیان ژنهای NRF-1 و NRF-2 کاردیومیوسیتها را افزایش دادند. همچنین نتایج نشان داد که بین این دو متغیر اثر تعاملی مثبت و معنیداری وجود دارد، به طوری که افزایش بیان ژنهای NRF-1 و NRF-2 در گروه تمرین + مکمل از سایر گروهها بیشتر بود. نتیجهگیری: به نظر میرسد هر دو عامل تمرین و مصرف مکمل کورکومین به تنهایی و در تعامل با هم میتوانند باعث افزایش بیان ژن برخی عوامل محرک بیوژنز میتوکندریایی در کاردیومیوسیت موشهای نر مبتلا به سکته قلبی شوند. همچنین، اثر تعاملی این دو عامل از اثرات جداگانه هر یک از آنها بیشتر میباشد.
Background& purpose: The aim of this study was to investigate the interactive effect of aerobic training and curcumin supplementation on mitochondrial NRF-1 and NRF-2 gene expression of cardiomyocytes in infarction model male rats. Methods: 32 male rats were randomly divided into four groups: control, aerobic training, supplementation, and aerobic training + supplementation. The training and training + supplementation groups participated in an aerobic training program for 8 weeks. The training was performed on an intelligent animal electronic treadmill 5 days a week, and at this time the control and supplementation groups did not have an training program. Also, curcumin supplementation was given to the supplementation and training + supplementation groups five days a week by gavage. After surgery, the expression of NRF-1 and NRF-2 genes in cardiomyocytes was obtained using real-time PCR method. Shapiro-Wilk and two-way analysis of variance were used for the analysis of data. Results: Both training and curcumin supplementation variables significantly increased the expression of NRF-1 and NRF-2 in cardiomyocytes. The results also showed that there is a positive and significant interaction between two variables, so that the increase in expression of NRF-1 and NRF-2 genes in the training + supplement group was more than the other groups. Conclusions: It seems that both training and curcumin supplementation alone and in interaction with each other can increase the gene expression of some factors that stimulate mitochondrial biogenesis in cardiomyocytes of male rats with myocardial infarction. also, the interaction effect of these two factors is greater than the separate effects of each of them.
