چکیده:
مقدمه: صنعت غذا در همه جای دنیا از اهمیت ویژهای برخوردار است. گوجه فرنگی و سایر فرآورده های آن نیز یکی از مهمترین محصولات کشاورزی در صنایع غذایی میباشند. گیاهان و بذرهای تراریخته یکی از دستاوردهای مهم بیوتکنولوژی نوین در زمینه کشاورزی هستند که در سالهای اخیر بخشی از بازارهای غذایی دنیا را تسخیر نموده اند. لذا شناسایی و بررسی تراریخته بودن یا نبودن محصولات کشاورزی مصرفی بدلیل اینکه هنوز نظرات موافق و مخالف متعددی برای استفاده از این محصولات در دنیا وجود دارد، امری ضروری میباشد.
مواد و روشها: در این پژوهش 10 نوع گوجه فرنگی به همراه کاسبرگ به روش نمونه گیری تصادفی ساده از بازار شهر تهران جمع آوری شد و مورد بررسی قرار گرفت. استخراج DNA انجام و به منظور تعیین غلظت و کیفیت DNA استخراج شده مقایسه هر نمونه با غلظت استاندارد DNA فاژ لامبدا بریده شده، با آنزیمهای برشی EcoRI، Hindǀǀǀ انجام شد. از آغازگرهای P35S F/R، NOS-1/NOS-3، RBCL-F/RBCL-R و NPTǁ-F/NPTǁ-R استفاده شد و قطعات نوکلئوتیدی بهترتیب bp195 و bp180 تکثیر شدند.
یافته ها: قطعات نوکلئوتیدی تکثیر شده شباهت 100% توالی نوکلئوتیدی را با نمونههای موجود در بانک ژنی تایید کردند که محصولات مورد بررسی به لحاظ ژنتیکی دست ورزی شدهاند. همچنین به منظور شناسایی گوجه فرنگیهای تراریخته، از جفت آغازگر PG F/R اختصاصی برای تکثیر آنتی سنس ژن پلی گالاکتوروناز، به کار رفت. تمام نمونه هایی که باند مربوط به پروموتور P35S یا خاتمه دهنده NOS یا هر دو را داشتند، باند bp384 مربوط به قسمتی از آنتی سنس ژن PG را تکثیر نمودند و شباهت 100% توالی ثبت شده ژن مذبور در بانک ژن محرز شد.
نتیجه گیری: نتایج این تحقیق نشان داد که گوجه فرنگی تراریخته در بازار ایران وجود دارد. اما به دلیل عدم برچسب گذاری به صورت گسترده، مصرف کنندگان از ماهیت تغییر یافته آنها اطلاع ندارند.
Introduction: Food industry has an important value all over the would. Tomatoes and it’s other products are one of the important agricultural products in food industry. Transgenic plants and seeds are one of the important achievements of modern biotechnology in field of agriculture that have captured a part of food marketing in recent years. Therefore, identify and examine whether transgenic agricultural products are transgenic or not, it is necessary because there are still many opinions for and against the use of these products in the world. Materials and Methods: In this study, 10 types of tomatoes along with sepals were collected by simple sampling method from Tajrish bazaar, Shahrvand-e farmaniyeh, vegetable market East and West of Tehran and were examined. At first, P35S F/R, NOS-1/NOS-3, RBCLF/R and NPTǁ F/R primers were used. Nucleotide fragments were amplified respectively at 195bp and 180bp. Results: Amplified nucleotide fragments confirmed 100% similarity of the nucleotide sequence to the samples in the gene bank that the studied products were genetically engineered. Also, in order to identify transgenic tomatoes, aspecific PG F/R primer pair was used to amplify the antigens of polygalacturonase gene. All specimens that had P35S promoter band or NOS terminator, or both, amplified the 384bp band associated with part of the PG gene antisense, and found a 100% similarity in the gene sequence registered in the gene bank. Conclusion: The results of this investigation showed that transgenic tomatoes exist in Iran’s market without consumer awareness.
